I. Aims
To observe the structure of the onion
cells and epithelial cells of the oral cavity
II.
Tools
and Materials
1. Microscope
equipped
2. Preparations glass / glass objects
3. Glass cover / deckglass
4. Pipette
5. Toothpick
6. Onion
7. Methylene blue
8. Epithelium of the oral cavity
III.
Procedures
1. Observations of plant cells in the
skin of red onion.
a. Prepare a microscope with amenities.
b. Make membrane
preparations of onion epidermis by peeling of the onion that
is still fresh by hand.
c. Put the epidermis / cuticle of onion with
just above a drop of water on a glass
object, then cover
with glass cover / deckglass.
d. Observe and draw on the tables that have been
provided.
e. To see the nucleus, you can give some methylene blue around the glass cover.
e. To see the nucleus, you can give some methylene blue around the glass cover.
f. Observe
and correct the result by drawing on teratur.
g. Measure the length of the object.
g. Measure the length of the object.
2. Observations on epithelial cells of
the oral cavity.
a. Take the inside of your cheek in the oral cavity by using a blunt toothpick carefully.
b. Put the scrapings on a glass preparations are already to add some water.
c. Stir with a toothpick and cover with cover glass.
d. Perform observations and measurements as in epidermal cells of onion and give some methylene blue to see the nucleus cells.

V. Questions
1. Mention
the form of onion cells and organel cell that looked
!
Answer : Nucleus, nucleolus,
mitochondria, chloroplast, ribosome, endoplasmic reticulum, golgi complex,
center vacuole.
2. Mention
the form of oral epithelial cells and organelles that looked!
Answer : Nucleus, nucleolus, mitochondria, endoplasmic reticulum, golgi complex,
lysosome, centriole..
3. Explain
the function of methylene blue !
Answer : Methylene blue serves to give color
organelles and cell organelles clarifying the observations (to see the nucleus
cell).
4. Write
the different between plant’s cells and animal’s cells of the observation
results !
Answer :
Different
:
ü Plant
cell larger than animal cells
ü Plant
cell have chlorophyll but animal not have chlorophyll
ü Plant
cell have cell wall but animal have no cell wall.
ü Plant
have a fixed shape but animal have no fixed shape.
5. Explain
the different and similarities between plant cells and animal cells, based on
literature which you read !
Answer :
Different
:
ü Plant
cell larger than animal cells
ü Plant
cell have chlorophyll but animal not have chlorophyll
ü Plant
cell have cell wall but animal have no cell wall.
ü Plant
have a fixed shape but animal have no fixed shape.
Similarities
:
ü Plant
cell have mitochondria and so animal cell.
ü Plant
cell have Reticulum Endoplasm and so animal cell.
ü Plant
cell have complex golgi and so animal cell.
ü Plant
cell have nucleus and so animal cell.
